Carlos Alcaraz is the defending champion.[1] This marked his first singles appearance in over four months, following a wrist injury he suffered at the Barcelona Open.[2]

Novak Djokovic's first-round defeat to Mariano Navone marked only his third career loss in the opening round of a major, his first opening-round loss at the US Open, and his earliest loss at a major since the 2006 Australian Open.[3] This marked the first time that none of the Big Three reached the second round of a major since the 2003 French Open.[4] As a result of his loss, Djokovic dropped out of the top 10 in the ATP rankings for the first time since July 2018, marking the first time since October 2002 in which none of the Big Three were ranked in the top 10.[5]
By defeating 12th seed Rafael Jódar, Bu Yunchaokete became the first lucky loser to defeat a top 15 seed in the first round of the US Open in the Open Era.[6][7][8]
This tournament marked the first absence from a major of current world No. 1 Jannik Sinner since the 2019 Wimbledon Championships.[9]
This also marked the final major appearances of 2016 champion, three-time major champion and former world No. 3 Stan Wawrinka, and former world No. 6 Gaël Monfils.[10] Wawrinka lost in the first round to Matteo Berrettini[11], while Monfils lost in the second round to Learner Tien.[12]
