フィラグリン

フィラグリン（Filaggrin）とは、表皮の顆粒細胞で産生される塩基性タンパク質の1種である^[1]。フィラグリンのことを指して、ヒスチジン・リッチ・プロテインなどと呼ぶ場合もある。ヒトにおいて皮膚のバリア機能に欠かすことのできない角質層を形成するに当たり、フィラグリンはケラチンと共に重要な役割を担っている。

フィラグリンは前駆体のプロフィラグリンとして生合成され、角質層が形成される段階で、リン酸プロフィラグリンが脱リン酸化と加水分解を受けて分解し、フィラグリンが作られる。プロフィラグリンは、フィラグリンが10個から12個繋がった巨大なタンパク質である。フィラグリンは、ケラチンと凝集・結合し、ケラチン・パターンを構成するが、最終的にはフィラグリンは角質上層でアミノ酸などの低分子まで分解される。
